The PacBio Revio long-read system leverages Single Molecule, Real-Time (SMRT) sequencing technology powered by Zero-Mode Waveguides (ZMWs). A laser illuminates the bottom of each ZMW, where a DNA polymerase is anchored; as fluorescently labeled dNTPs are incorporated, they emit distinct signals that enable precise base identification. The system operates in Circular Consensus Sequencing (CCS) mode—generating HiFi reads by combining multiple subreads from the same DNA molecule into a single consensus sequence. This delivers exceptional accuracy with quality values exceeding Q30 (>99.9%).
