Introduction
Human Platelet Antigen (HPA) genotyping is a pivotal diagnostic procedure in transfusion medicine, hematology, and organ transplantation. The HemoSure™ Long-Read Sequencing HPA Genotyping Kit (Single-Molecule Real-Time Sequencing) leverages PacBio HiFi long-read sequencing technology to deliver high-resolution genotyping. It provides precise coverage of HPA loci 1 through 35, encompassing seven core genes: CD109, ITGA2B, GP1BB, GP9, ITGA2, ITGB3, and GP1BA
This kit is designed for the qualitative detection of HPA-related genes in whole blood samples. It enables high-resolution genotyping of HPA loci 1 through 35, covering seven specific genes: CD109, ITGA2B, GP1BB, GP9, ITGA2, ITGB3, and GP1BA. The assay provides auxiliary diagnostic support for clinical scenarios including platelet transfusion matching, diagnosis and treatment of hematological diseases, and donor-recipient screening for organ transplantation.
The workflow utilizes polymerase chain reaction (PCR) to amplify a predefined set of target genes. The resulting amplicons are designed for downstream library preparation and sequencing on PacBio HiFi platforms. The generated sequencing data is subsequently analyzed using Haorui TransMatchTM software to produce high-resolution HPA genotyping results.
Note: Test results obtained with this kit are for clinical reference only and should not be used as the sole basis for diagnosis or clinical decision-making. Clinicians should make comprehensive judgments and treatment decisions by integrating these results with the patient’s medical history, physical examination, and other laboratory findings.
HemoSure™ Long-Read Sequencing HPA Genotyping Kit serves as an integrated solution for amplifying target genes of the HPA and preparing sequencing libraries. The kit facilitates the amplification of CD109, ITGA2B, GP1BB, GP9, ITGA2, ITGB3, and GP1BA genes, followed by library preparation for downstream sequencing.
(1) Nucleic Acid Extraction: Isolate DNA from samples.
(2) PCR Enrichment: Amplify target genes (CD109, ITGA2B, GP1BB, GP9, ITGA2, ITGB3, and GP1BA).
(3) Library Prep: Ligate PacBio’s proprietary SMRTbell adapters to construct circular DNA libraries.
(4) Sequencing: Load libraries onto Revio’s or Vega’s long-read sequencing platform with HiFi sequencing mode.
(5) Fluorescently labeled nucleotides are incorporated during polymerization, and real-time signals are captured to generate HiFi reads.
(6) Data Analysis: Align reads to the human genome and CD109, ITGA2B, GP1BB, GP9, ITGA2, ITGB3, and GP1BA reference sequences and export the HPA genotyping results.

Shipping and Storage
1. Storage Conditions
Store the kit at -20 ±5°C. The shelf life is 3 months from the date of manufacture (Note: Magnetic beads should be stored in a 4°C refrigerator).
2. Freeze-Thaw Stability
The kit may undergo no more than 6 freeze-thaw cycles. Once opened, use immediately.
3. Shipping Requirements
Transport under temperature-controlled conditions (-20 ±5°C).
Maximum allowable transit time: 3 days.
Upon receipt, verify that the internal temperature remains within -20 ±5°C before opening.
4. Refer to the lot-specific label for manufacturing date (MFG) and expiration date (EXP).
